摘要 本研究以棉花干种子为材料,探索一种适用棉花品种SSR分析的DNA快速提取方法。先采用SDS提取液进行一步裂解,并将RNA的去除操作融入抽提过程,从而简单快速的获得高质量棉种基因组DNA。紫外检测与琼脂糖凝胶电泳检测结果均表明,所得DNA纯度高,完整性好。另外,SSR分析结果也显示,扩增带型清晰易辨,重复性与稳定性好。相比常规的以棉花幼苗叶片为材料的DNA提取法,本方法在降低实验成本的同时大大提高了工作效率。 We applied single dry cotton seed.to be as materials for exploring a rapid method of DNA extraction appropriate for SSR analysis in cotton variety.In the method,just one step was used for cell lysis by SDS buffer,and the operation of RNA digestion was integrated into protein extraction,then high quality genomic DNA from cotton seed was obtained simply and quickly.In this paper the results of UV detection and agarose gel electrophoresis all showed that the purity and integrity of the isolated DNA were satisfying.In addition,as the results of SSR analysis,the amplified bands were clear as well as good in repeatability and stability.Compared to conventional leaf DNA extraction,the method based on seed reduced cost as greatly improved work efficiency.
