摘要
从转基因棉花后代分离群体中选择含有外源基因的目标植株是培育转基因棉花的重要环节。为了快速在早代对转基因棉花进行有效的筛选,本试验利用含有Kan的水溶液在培养皿上对转基因棉花YZ1进行筛选研究。结果表明,100mg/L的Kan能有效抑制非转基因棉花种子的萌发及下胚轴的伸长;经PCR检测,100mg/LKan筛选出的抗性苗阳性率达到95%以上。
To screen target plants containing foreign genes from the segregated generations of transgenic cotton is an important part for developing transgenic cottons.In order to screen rapidly and effectively in the early generations of transgenic cotton.In this study,screening of transgenic cottons was researched with aqueous solution containing Kanamycin in the petri dish.The results showed that the germination of non-transgenic cotton seeds and hypocotyl elongation were inhibited in the solution of 100 mg /L Kan.The positive rate was more than 95% by PCR detection of 100 mg /L Kan resistant seedlings.
