依达拉奉对脓毒症大鼠心肌线粒体SOD XOD及心肌组织NF-κB的影响 被引量:4

摘要 目的 观察依达拉奉对脓毒症大鼠心肌线粒体超氧化物歧化酶(SOD)、黄嘌呤氧化酶(XOD)及心肌细胞NF-κB表达的影响.方法 SD大鼠30只分为假手术组、手术对照组、依达拉奉治疗组,手术对照组和治疗组均予盲肠结扎穿刺法制作大鼠脓毒症模型,3组术前15 min及术后3 h各皮下注射盐酸左氧氟沙星20 mg/kg,治疗组于术前15 min及术后3 h各皮下注射依达拉奉5 mg/kg,3组术后18 h取心肌线粒体测SOD、XOD,及用RT-PCR法测定大鼠心肌组织的NF-κB mRNA的表达,并对心肌线粒体进行电镜检查.结果 手术对照组心肌细胞线粒体SOD活力显著低于假手术组,心肌组织NF-κB mRNA的表达显著高于假手术组;治疗组心肌细胞线粒体SOD活力显著高于手术组,心肌组织NF-κB mRNA的表达显著低于手术组,各组间心肌细胞线粒体XOD活力比较差异无统计学意义.手术对照组电镜下可见心肌线粒体轻度水肿,治疗组心肌线粒体嵴多、致密(正常).结论 依达拉奉能使心肌线粒体SOD活力升高,并使心肌细胞NF-κB mRNA的表达下降,提高清除自由基和抗氧化能力,减轻炎症级联放大效应,具有保护心肌作用. Objective To observe the effect on superoxide dismutase (SOD), xanthine oxidase (XOD) of mitoehondria in myocardium and NF - κB in myoeardium of sepsis rats treated with edaravone. Methods 30 Sprague - Dawley rats were divided into 3 grouds : sham operated group( group A) , controlled operated group( group B) , treated group with edaravone (group C ). The model of sepsis rats were made by the way of caecum ligated and punctured(CLP) and 20 mg/kg lactate levofloxaein was subcutaneously injected(sci) 15 min before and 3 h after operation in three group. 5 mg/kg edaravone were subcutaneously injected(sci) 15 min before and 3 h after operation in group C. Mitoehondrias in myoeardium were taken from all of them 18 h after operation. The activities of SOD, XOD were detected of mitoehondria in myoeardium. The expression of NF - κB mRNA in myoeardium tissue was evaluated by RT - PCR . Pathological change of myocardium were observed. Results The activities of SOD in group B were significantly decreased compared group A. The activities of SOD in group C were significantly increased compared group B. The expression of NF - κB mRNA in group B were significantly increased compared group A. The expression of NF - κB mRNA in group C were significantly decreased compared group B. There were no significant difference among the three groups in XOD. The mitochondria in myocardium was swelling by electron microscope in group B. But it was normal in group C. Conclusion The activities of SOD of mitochondria in myocardium increased and expressionof NF - κB mRNA in myocardium tissue decrease in sepsis rats while treated with edaravone, which could enhance ability of antioxidating and scavenging free radical , inflammatory cascade, protect Cardiac function.
