盐酸戊乙奎醚对吗啡依赖大鼠条件性位置偏爱重现效应的影响 被引量:3

摘要 目的 评价盐酸戊乙奎醚对吗啡依赖大鼠条件性位置偏爱重现效应的影响.方法 雄性成年SD大鼠40只,体重180~220 g,随机分为5组(n=8):对照组(C组)、吗啡依赖组(M组)和不同剂量盐酸戊乙奎醚组(P1-3组).M组和P1-3组连续8 d交替皮下注射吗啡10 mg/kg(9:00)或生理盐水(16:00),诱导大鼠条件性位置偏爱效应;然后连续皮下注射生理盐水(9:00和16:00)10 d,使形成的条件性位置偏爱效应逐渐消退;单次引燃皮下注射吗啡4 mg/kg,诱导消退的条件性位置偏爱效应重现;C组用生理盐水替代.在单次引燃皮下注射吗啡前30 min时,P1-3组腹腔注射盐酸戊乙奎醚0.5、1.0、1.5 mg/kg,C组和M组腹腔注射等容量生理盐水.所有处理结束后进行条件性位置偏爱测试,记录大鼠15 min内在灰区停留的时间.结果 与M组比较,P1-3组灰区停留时间缩短(P<0.05或0.01).与P1组比较,P2组灰区停留时间差异无统计学意义(P>0.05),P3组灰区停留时间缩短(P<0.05).结论 盐酸戊乙奎醚可抑制吗啡依赖大鼠条件性位置偏爱重现效应,且与剂量有关. Objective To investigate the effect of penehyclidine hydrochloride (PHCD) on the reinstatement of conditioned place preference (CPP) in morphine dependent rats. Methods Forty male adult SD rats weighing 180-220 g were randomly divided into 5 groups (n = 8 each): group control (group C); group morphine (group M) and 3 PHCD groups (group P1-3 ). Morphine 10 mg/kg was injected subcutaneously once a day for 8 days to induce morphine CPP. The rats were then subjected to extinction of CPP for 10 days with normal saline (NS) instead of morphine. After the extinction, the rats were put into the drug-paired side of the box. A single priming dose of morphine 4 mg/kg was injected to reinstate the morphine CPP. In group P1-3 the rats received PHCD 0.5, 1.0 and 1.5 mg/kg intraperitoneally 30 min prior to priming dose of morphine, whereas in group C and M the rats received NS. The second day the rats underwent CPP test. Results Compared with group M, the time spent in the drug-paired side (grey area) was significantly shortened in group P1-3 (P 〈 0.05 or 0.01 ).Compared with group P1 ,no significant change in the time spent in the drug-paired side (grey area) was found in group P2(P 〉 0.05), but the time spent in the drug-paired side (grey area) was significantly shortened in group P3 ( P 〈 0.05). Conclusion PHCD could significantly inhibit the reinstatement of CPP induced by priming dose of morphine in morphine dependent rats and it is related to the dose.
