重楼总皂苷对人肺癌细胞A549的增殖抑制作用及对细胞周期的影响

Effect of steroidal saponins from P. polyphylla on proliferation and cell cycle of A549 cells

目的:研究重楼总皂苷对人肺癌细胞A549的增殖抑制作用及对细胞周期的影响。方法:用MTT法测定重楼总皂苷对A549细胞的增殖抑制率;Hoechst-PI双染观察细胞形态学变化;流式细胞术测定重楼总皂苷对A549细胞周期时相分布的影响。结果:重楼总皂苷能抑制A549细胞的增殖,其作用呈明显的时间和剂量依赖性;Hoechst-PI双染显示细胞凋亡特征;重楼总皂苷使A549细胞被阻滞在S期。结论:重楼总皂苷对A549细胞的增殖有较强的抑制作用,明显影响细胞周期时相分布。

Objective To study the effect of steroidal saponins from P. polyphylla on proliferation and cell cycle of A549 cells. Methods The inhibition rate of steroidal saponins from P. polyphylla for A549 cell proliferation was detected by MTT. The morphological changes were determined using Hoechst-PI staining. The effect of steroidal saponins from P. polyphyUa on the cell cycle of A549 was observed by flow cytometry. Results Steroidal saponins from P. polyphyUa inhibited the cell proliferation of A549 cells in a dose- and time-dependent manner. Hoechst-PI staining showed the features of apoptosis. A549 cells treated by steroidal saponins from P. polyphylla were blocked in S phase. Conclusions Steroidal saponins from P.polyphylla can inhibit the growth of A549 cells by inducing S phase arrest.
