hGPR 87基因表达与宫颈癌发生及浸润转移的关系

摘要 目的:通过检测宫颈癌及宫颈上皮内瘤变(CIN)患者宫颈组织hGPR87基因表达水平来分析基因表达与宫颈癌发生及浸润转移的关系。方法:采用SYBRGreenI实时定量PCR方法分别检测30例宫颈癌、30例重度宫颈上皮内肿瘤(HCIN)、30例轻度宫颈上皮内肿瘤(LCIN)患者和15例正常对照(子宫肌瘤)新鲜宫颈组织的hGPR87基因表达情况,以β2微球蛋白基因(β2M)作为内参,根据相对定量公式(2-△Ct×100%)计算hGPR87基因相对表达量。结果:宫颈癌患者组的表达量最高,HCIN组次之,正常对照组的表达量最低,四组比较差异有统计学意义(P=0.02)。宫颈癌患者中有淋巴结转移的比无淋巴结转移的宫颈组织中hGPR87的mRNA含量要高(P=0.002)。结论:本研究揭示了不同宫颈疾病发展阶段hGPR87基因表达的变化特点,hGPR87基因表达水平在宫颈癌及伴淋巴结转移患者以升高为主,可能与宫颈癌发生及浸润转移有一定关系。 Objective To investigate the relationship between hGPR 87 mRNA expression and cervix carcinoma. Methods SYBR green Ⅰ real-Time PCR was used to detect hGPR 87 mRNA expression in cervical tissue from 30 cases of cervix carcinoma, 30 cases of high-grade cervical intraepithelial neoplasia, 30 cases of low-grade cervical intraepithelial neoplasia and 15 healthy individuals. β2-microglobulin (β2M) gene was used as an internal control. The relative expression of hGPR 87 mRNA was analyzed using the 2-Δα method. Results hGPR 87 mRNA level is the highest in cervix carcinoma patients and the lowest in healthy individuals,with statistical difference (P = 0.02). hGPR 87 mRNA expression between cervix carcinoma patients with and without lymph node metastasis is also significantly different (P = 0.002). Conclusions This is, to our knowledge, the first report about hGPR 87 mRNA expression feature in cervix carcinoma patients.hGPR 87 mRNA expression was increased in most of cervix carcinoma patients, which may contribute to tumorigenesis and metastasis of cervix carcinoma.
