一种新的红细胞生成调控因子P59蛋白质的分离纯化和性质研究

摘要 以正常人尿为原料,经过DEAE纤维素离子交换层析、硫酸铵沉淀、凝胶过滤、碱性制备电泳等纯化步骤得到一个分子量为59kDa的蛋白质,称为P59蛋白.该蛋白质在体外可以促进13d胎鼠肝细胞红系集落的形成,在体内可以刺激小鼠网织红细胞的生成,促使5/6肾切除大鼠贫血症状和Hymann肾炎大鼠贫血症状的改善,对尿毒症病人的贫血症有明显疗效. A novel protein (P59 protein) with mol wt 59kDa has been found widely present in serum, urine and kidney cell conditioned medium. The protein possesses the same biological activity as erythropoietin(EPO), in stimulating the CFU E formation in vitro and correcting the anemia of animal models in vivo. P59 protein also corrects the anemia of uremic patients effectively. In this paper, a P59 protein purified from urine of young men is reported. The purification procedures include DEAE cellulose chromatography, (NH 4) 2SO 4 precipitation, sephacryl S 200 gel filtration and preparative gel electrophoresis. The purified final product shows a single band when run on SDS PAGE and a single peak is observed by HPLC analysis. The N terminal sequence of this P59 protein is quite different from all known proteins. According to its biological activity in vitro and in vivo, the results of N terminal amino acid sequence analysis and dot ELISA, P59 potein proves to be a novel factor, which can play an important role in the regulation of erythropoiesis.
