苦参类生物碱对HBV转基因小鼠抗病毒作用的比较研究

Comparative study on antiviral action of Ganyanling injection,Kushensu injection and Kushensu capsule in HBV transgenic mice

目的比较研究肝炎灵、苦参素注射液和苦参素胶囊的抗乙型肝炎病毒(HBV)作用。方法M-TgHBV转基因小鼠38只,随机分为5组,生理盐水组、拉米夫定组、苦参素胶囊组分别按20.59mg.kg-1.d-1、123.3mg.kg-1.d-1予以灌胃;肝炎灵注射液组、苦参素注射液组分别按14.43mg.kg-1.d-1、82.2mg.kg-1.d-1,予以腹腔注射,共28d;应用酶联免疫吸附法(ELISA)检测HBsAg、HBeAg含量,应用实时荧光定量聚合酶链反应(PCR)法检测HBVDNA含量。结果对HBsAg表达的抑制作用,生理盐水组与拉米夫定组间以及苦参素注射液与肝炎灵注射液组间比较没有统计学差异,P>0.05,其他各组之间均有显著性差异,P<0.01;对HBeAg的抑制作用,生理盐水与拉米夫定组间以及苦参素注射液组与苦参素胶囊组之间差异无统计学差异,P>0.05,其他各组之间比较均有显著性差异,P<0.01;各组之间对HBVDNA的抑制作用比较均有显著性差异,P<0.01。结论肝炎灵注射液、苦参素注射液以及苦参素胶囊均有一定抗HBV作用,抗病毒能力由强到弱依次是肝炎灵注射液、苦参素注射液、苦参素胶囊。

Objective To comparatively study antiviral effects of Ganyanling injection,Kushensu injection and Kushensu capsule.Methods Thirty-eight M-Tg HBV transgenic mice were randomly divided into five groups.Physiologic saline group,lamivudine group,Kushensu capsule group were respectively intragastricly administrated at dosage of 20.59 mg·kg-1·d-1,123.3 mg·kg-1·d-1 for 28 days;Ganyanling injection group and Kushensu injection group were respectively administrated at dosage of 14.43 mg·kg-1·d-1,82.2 mg·kg-1·d-1 by intraperitoneal injection for 28 days.The levels of HBsAg and HBeAg in the liver were detected by an enzyme-linked immunosorbent assay（ELISA）.The levels of HBV DNA in the liver were detected by a quantitative real-time polymerase chain reaction（RT-PCR）.Results In repression of HBsAg expression,no obvious change was seen in saline group-to-lamivudine group and Kushensu injection-to-Ganyanling injection（P0.05）;significant difference were seen between other groups（P0.01）.In repression of HBeAg,no obvious change was seen in saline group-to-lamivudine group and Kushensu injection-to-Kushensu capsule（P0.05）;Significant differences were seen between other groups（P0.01）.There was significant difference between each group in repression of HBV DNA.Conclusions Ganyanling injection and Kushensu injection and Kushensu capsule all exert definite repressive effects on the HBV.The powerful action is orderly Ganyanling injection,Kushensu injection,Kushensu capsule.
