摘要
目的探讨胃癌组织中hTERT、hMSH2基因在胃癌发生中的作用和Hp致癌的可能机制。方法采用W-S法检测93例胃癌患者癌组织及癌旁组织中Hp感染情况;利用免疫组织化学方法检测93例胃癌患者癌组织及癌旁组织中hTERT、hMSH2的表达情况。结果(1)胃癌组织hTERT阳性率为79.6%,明显高于癌旁组织22.6%(P<0.01);Hp感染组hTERT蛋白表达阳性率高于非感染组(P<0.01);(2)胃癌组织hMSH2阳性率为65.6%,明显高于癌旁组织38.7%(P<0.01);Hp感染组hMSH2基因蛋白表达阳性率低于非感染组(P<0.05)。结论Hp诱导的端粒酶的表达增高和错配修复基因hMSH2的表达异常可能与胃癌的发生有关。
Objective To explore the relationship between the expression of hTERT and mismatch repair gene hMSH2 in gastric cancer and the possible mechanism of carcinogenesis caused by Hp infection.Methods Hp infection was determined with W-S method in 93 gastric cancer and adjacent mucosa tissues.Immunohistochemistry SP method was used to examine the expression of hTERT and hMSH2 in gastric cancer and adjacent mucosa sections.Results The positive rate of hTERT in the gastric cancerous lesions was 79.6%,which is significantly higher than that in the neighboring normal mucosa(22.6%)(P0.01).The hTERT positive rate in the Hp positive patients was higher than the Hp negative patients(P0.01).The positive rate of hMSH2 in the gastric cancerous lesions was 65.6%,which is significantly higher than that in the neighboring normal mucosa(38.7%)(P0.01).The hMSH2 positive rate in the Hp positive patients was lower than the Hp negative patients(P0.05).Conclusions Hp induced changes in the expression of telomerase and high expression of hMSH2 may be associated with the development of gastric cancer.
