摘要
本研究以溆浦甜橙实生苗为试材,对影响其离体再生的各种因素进行了研究,建立了一个高效离体再生系统。结果表明,30 d龄溆浦甜橙实生苗上胚轴水平置于MT+3 mg·L-1 6-BA分化培养基中,不定芽的诱导率可高达83.3%,该不定芽在MT+0.5 mg·L-1 6-BA+1.5 g·L-1麦芽提取物的增殖培养基上生长良好,增殖率可达96.7%,增殖系数为3.79。生根培养基以1/2MT+0.5 mg·L-1 IBA比较适宜。这一系统再生频率高,操作简便,适合于溆浦甜橙的遗传转化。
Using epicotyls of 30 d seedlings of sweet orange (Citrus sinensis Osbeck Xupu)grown in vitro as explants, an efficient regeneration system with a high frequency was established. The best results were obtained when epicotyls were placed horizontally on the surface of culture medium. The optimal induction bud medium was MT containing 3 mg·L^-1 6-BA and bud induction reach 83.3 %. The shoot multiplication medium was MT containing 1.5 g·L^-1 malt extracted and 0.5 mg·L^-1 6-BA and muhiphcation rate, coefficient reach 96.7 %, 3.79 respectively. The highest rooting percentage was obtained on 1/2MT medium containing 0.5 mg·L^-1 IBA. This system is high regeneration rate, convenient operation and suitable to genetic transformation.
