全反式维甲酸对鼻息肉术后囊泡上皮细胞PCNA和FasL表达的影响 被引量:1

摘要 目的探讨全反式维甲酸(ATRA)对鼻息肉术后术腔组织增殖细胞核抗原(PCNA)及FasL表达的影响。方法鼻息肉手术患者50例,随机分为ATRA组25例(术后采用ATRA治疗)和囊泡组25例;另选择鼻中隔偏曲矫正手术患者下鼻甲组织15例作为对照组。取三组患者鼻黏膜上皮标本经石蜡包埋,HE染色,采用免疫组化方法行PCNA和FasL蛋白的检测。结果ATRA组、囊泡组、对照组PCNA蛋白表达的光密度值分别为1.61±0.2、4.84±0.2、2.41±0.2,FasL蛋白表达则分别为3.35±0.2、6.56±0.2、2.16±0.2。ATRA组与囊泡组比较,PCNA及FasL蛋白表达差异有统计学意义(P<0.01或<0.05)。增生组织上皮细胞PCNA蛋白与FasL蛋白表达呈明显的正相关(r=0.834,P=0.011)。结论ATRA可以抑制鼻息肉术后囊泡上皮细胞的增殖活性,该机制可能与其下调上皮细胞FasL表达,进而诱导了囊泡上皮细胞的凋亡有关。 Objective To investigate the effects of all-trans retinoic acid(ATRA) on expression of PCNA and FasL in the epithelia of vesicle tissues after endoscopic polypectomy.Methods A total of fifty patients with nasal polyps after endoscopic sinus surgery was divided into two groups randomly,namely ATRA group(25 patients,treated with ATRA),vesicle tissues group(25 patients),and 15 normal inferior turbinate cases in control group were included in this study.All specimens were imbedded by paraffin,stained by HE.The protein expressions of PCNA and FasL were detected by immunohistochemical staining(PV 9000 two-step kit).Results For the positive expression of PCNA,the average density of light in epithelia of ATRA group,vesicle tissues group and control group were 1.61±0.20,4.84±0.20 and 2.41±0.20 respectively.And for FasL,they were 3.35±0.20,6.56±0.20,2.16±0.20 respectively.The expression of PCNA and FasL in the epithelia of ATRA group and vesicle tissues group were statistically significant(P0.01 or 0.05).In the epithelia of hyperplasia tissues,it displayed positive correlation between the expression of PCNA and FasL(r=0.834,P=0.011).Conclusions ATRA can inhibit the proliferative ability of epithelia in the vesicle tissues after polypectomy.The mechanism for ATRA restraining epithelial proliferation may be related with the down-regulating the expression of FasL,which induced apoptosis of the epithelia in vesicle tissues.
