摘要
对本实验室提取的川产天麻多糖粗提液进行分离纯化,研究结果表明:采用酶法除蛋白效果比较理想,一次就能达到比较好的效果,胰蛋白酶用量为2.0U/mg,处理时间以5h最好;酶解结合一次Sevag法再醇沉后,蛋白脱出率为90.1%;进样体积8ml、样品浓度4%、流速0.8ml/min时,Sepharose 6 Fast Flow凝胶分离效果最好。
The results of methods（Sevag,enzyme and enzyme decomposition with Sevag）showed that：Enzyme method is the best one to eliminate protein with dosage of enzyme 2.0U/mg for 5 hours.Enzyme decomposition with Sevag,then precipitated with ethanol.Remove rate of protein is 90.1%。The separation conditions of Sepharose 6 Fast Flow gel are 8ml 0f sample volume,4% of samples concentration 4%,and the velocity of 0.8ml/min.
