摘要
目的:体外探讨肿瘤坏死因子-α(TN F-α)和白细胞介素-6(IL-6)对关节软骨细胞胶原表型表达的影响。方法:分离成人正常关节软骨细胞,用 TN F-α单独或与IL-6联合干预培养,采用形态学观察、流式细胞仪分析、Nort hern杂交技术,检测关节软骨细胞表型。结果:TNF-α可促使软骨细胞向成纤维细胞样细胞形态转化及分裂、增殖,抑制其表达特异性Ⅱ型胶原,而诱导其表达非特异性Ⅲ型胶原;IL-6有协同 TN F-α干扰软骨细胞表型表达的作用。结论: TNF-α和IL-6是参与关节软骨细胞变性的介导因素。
Objective:To reveal the regulating effects of on the phontypical expression of articular chondrocytes cultured with tumor necrosis factor-a (TNF-a)and interleukin-6(IL-6). Method: Chonforytes of human normal articular cartilage obtained for young adults died of traffic accident were cultured. The characteristics of chondrcyte morphology was observed with time-lapse microvideo-recorder attached to phase contrast micro- scope, and hitochemistry,The expression of procollage collagen Type Ⅰ, Ⅱ, Ⅲ mRNAs was detected with northern blot technique.Results: Mor- phology of articular chondrocytes cultured with TNF-α tranfered to fibroblasts-like cells;TNF-α could suppress the cartilagespecific Ⅱprocolla- gen gene expression and induce expression of cartilage-nonspecific Type Ⅲ mRNA in the early culture phase of human artilar chondrocytes. TNF-α in combination with IL-6 could obviously interfere the phontypical expression of chondrocfes in the late culture phases.Conclusion:TNF- α and IL-6 are important factors in the control of chondrocytes differentiation.
