摘要
研究了表达酪氨酸酶的基因工程菌(E.coliWY8)产黑色素的条件。WY8 能在酪素平板上产生可溶性黑色素,在添加酪氨酸的液体LB培养基中的黑色素产率是基因来源菌—Pseudom onasm altophilia 的5 倍。用正交试验法找到适宜发酵产黑色素的培养基,对摇瓶装量、菌龄、底物浓度、发酵温度和pH 等因素的影响作了进一步探讨。
The condition for the production of melanin by engineering bacteria (E.coli WY8) bearing tyrosinase gene was examined.WY8 could form soluble melanin pigments on the casein agar plates.Its melanin productivity was 5 times as much as the bacteria of tyrosinase gene source (Pseudomonas maltophilia) in liquid LB culture supplemented with tyrosine.By means of orthogonal transformation the suitable medium for melanin fermentation was obtained.The effects of flask volume,culture age,substrate concentration,fermentation temperature and pH were also studied.
