摘要
建立了奥美拉唑体内含量测定方法。具有内标物硝苯地平易得、流动相成本较低、操作简便、灵敏度高等优点,且奥美拉唑的2个代谢物可以完全分离。
The RP HPLC method for determination of omeprazole (1) and its two metabolites in plasma and for study on oral pharmacokinetics of 1 was established. A stainless steel column packed with YWG C 18 (3.9×150 mm, 10 μm) was used. The mobile phase was methanol acetonitrile pH8 buffer(45∶10∶45, v/v ) with flow rate of 1.0 ml·min -1 at room temperature. The detector was set at 302 nm. Calibration curve was linear ( r =0.9999) in the concentration range of 10~1500 ng·ml -1 . The within day and between day precision ( RSD ) were less than 9% with average recoveries of 99.6%~ 102.8%.
