摘要
目的:研究化疗药物长春新碱(Vincristine,VCR)对人肺癌细胞株A549生长的抑制作用以及Livin基因表达的变化。方法:将VCR按100、50、25、12.5、6.25、3.13、1.56、0.78μg/ml浓度加入A549后,分别作用24、48、72小时,用噻唑蓝(MTT)比色法检测细胞增殖抑制率;实时荧光定量PCR检测LivinmRNA的表达,免疫印迹法(Westernblot法)检测Livin蛋白的变化。结果:VCR对A549具有一定增殖抑制作用,呈时间和剂量依赖性。VCR早期可抑制A549中Livin基因表达,作用72h后Livin基因表达量开始增加。结论:VCR可抑制A549肺癌细胞的增殖,在一定时间内呈剂量、时间依赖性。VCR可能诱导了Livin基因的表达,Livin可能参与了肺癌细胞对VCR的耐药机制。。
Objective: To investigate the inhibition of Vincristine (VCR) on Livin gene expression in human lung cancer cell line (A549) in vitro. Methods: The proliferation inhibition ratio was evaluated by MTT in the time of 24, 48 and 72 hours. The adopted inhibiting concentration (IC50) of VCR was added into the medium of A549 cells. The expression of Livin gene was detected by real time PCR, and the protein of Livin was assessed by Western-blot. Results: The proliferation of A549 was inhibited in a dose- and time-dependent manner after the treatment of VCR. The expression of Livin was down-regulated in the earlier period, and was up-regulated after 72 hours by VCR. Conclusion: VCR can inhibit the proliferation of A549 in a dose- and time-dependent manner within a time range. VCR can induce the expression of Livin gene in lung cancer cells,which may participate in the drug resistance in someway.
