摘要
目的:探讨整合素α6在宫颈癌中的表达及意义。方法:在20例宫颈浸润癌中整合素α6基因及其蛋白分别采用反转录聚合酶链反应(RT-PCR)、免疫组织化学(IHC)SP法进行检测。18例正常宫颈上皮、20例宫颈原位癌、55例宫颈浸润癌中(包括前20例宫颈浸润癌),整合素α6蛋白的表达采用免疫组织化学SP法检测。结果:20例宫颈浸润癌中整合素α6mRNA和蛋白表达率无统计学差异(P>0.05)。整合素α6在宫颈癌中的表达率明显低于正常宫颈组织(P<0.001),而宫颈浸润癌和宫颈原位癌组织之间无显著性差异(P>0.05)。整合素α6表达与宫颈鳞癌临床分期、肿瘤大小、病理分级、宫颈浸润深度无相关(P>0.05),而与淋巴转移相关(P<0.05)。结论:整合素α6可能通过改变宫颈癌细胞的粘附力而发挥促进宫颈癌发生发展及淋巴结转移的作用。
Objective: To study the expression and significance of integrin alpha 6 in cervical cancer. Methods: Integrin alpha 6 mRNA and its protein were examined by reverse transcription polymerase chain reaction (RT-PCR) immunohistochemical(IHC) SP method respectively in 20 cases of invasive cervical cancer.The expression of integrin alpha 6 protein was tested by IHC SP method in 18 cases of normal cervical epithelium,20 cases of carcinoma in situs of cervix and 50 cases of invasive cervical cancer(including the former 20 cases of invasive cervical cancer). Results: There was no significance between expression of integrin alpha 6 mRNA and its protein (P0.05).The expression of integrin alpha 6 protein in cervical cancer was obviously decreased than normal cervical tissue (P0.001),and there was no statistics significance between invasive cervical cancer and carcinoma in situs of cervix (P0.05) .There was no relation between the expression of integrin alpha 6 protein and clinical stage,size of tumor,pathological classification and the depth of invasive cervical (P0.05),but it was ralated with lymph node metastases (P0.05). Conclusion: Integrin alpha 6 may change cell adhersion of cervical cancer and promote development, progress and metastasis of this cancer.
