摘要
感染小鼠淋巴结 B细胞与小鼠骨髓瘤 SP 2/0株融合,所获单克隆抗体 ⅢD10能与血吸虫抗原形成免疫沉淀,具有属特异性,免疫荧光定位于成虫肠管。免疫电泳提示,ⅢD10为抗循环阴极抗原单抗。应用于竞争 ELISA试验,显示很高的敏感性和特异性,检测 120例烘检阳性慢性血吸虫病患者,100％阳性,204例正常人仅1例在1:10时出现阳性,与6例肺吸虫病人无交叉反应。感染动物试验,感染前30只家兔均阴性,感染后4周全部出现阳性;治疗后8周抑制滴度逐渐下降。
A monclonal antibody IIIDIO was obtained by hybridization of B lymphocyfes from mice infected with S. japonicum, and SP 2/o myeloma cells. An important characteristic of the McAb IIIDIO is its capacity to immunoprecipitate schistosome antigens. A genus specificity for schistosome was found. IF A test showed that positive fluorescence reaction was located on the caecum of adult worms. A single cathodic precipitation line was revealed by immunoelectrophoresis.Competitive ELISA shown that a positive rate of 100% in 120 sera of chronic patients with positive stool examination, and only one false positive in 204 healthy individuals were obtained In infected rabbit models, the inhibitory titer declined progressively after praziquantel treatment.
