单克隆抗体斑点试验检测血吸虫卵抗原 被引量:9

DETECTION OF SCHISTOSOME OVA ANTIGEN BY DOT-ELISA WITH MONOCLONAL ANTIBODY

摘要 靶抗原为血吸虫卵糖蛋白的单克隆抗体SM21-3标记过氧化物酶后,用于直接法斑点试验检测血清中虫卵抗原。12份感染兔血清全部呈阳性反应;12份正常兔血清呈阴性反应。8份感染 10 0条及8份感染10条尾蚴的治疗前兔血清呈阳性反应;治疗后1月,血清抗原水平有不同程度下降;治疗后3月,部分血清转为阴性,其余几份均呈弱阳性反应。检测血吸虫病人血清80份,阳性66份,阳性率为82.5%;检测正常人血清60份,未见阳性反应。上述结果表明本方法具有较好的敏感性与特异性,而且方法简单易行,肉眼观察结果,适于现场应用。 The monoclonal antibody SM21-3 (against egg glycoprotein of Schistoaoma japonic-um) labelled with peroxidase was used in Dot-ELISA test for detecting egg antigen in serum. Positive reactions were found in specimens of serum from 12 rabbits infected with Schistosoma japonicum and negative in 12 normal rabbits. 8 rabbits infected with 100 cercariae and 8 rabbits infected with 10 cercariae were all positive before treatment. One month after treatment, antigen levels in sera decreased in different degrees. Three months after treatment, some of them turned negative, others showed only weakly positive reactions. 80 specimen serum from patients from Anhui and Hubei were examined, 66 were positive. The positive rate was 82.5%. None was positive in 60 normal individuals. The above results showed that the sensitivity and specificity of direct Dot-ELISA with monoclonal antibody were high enough for detecting the egg antigen. Since the method is simple and easy and the result is readable with naked eye, it can be used in any field station.
