摘要
将中国番茄黄化曲叶病毒(tomato yellow leaf curl China virus,TYLCCNV)外壳蛋白基因(coat protein,CP)同源的部分片段双链RNA重组至植物表达载体pBIN19,通过根癌农杆菌(Agrobacterium tumefaciens)菌株LBA4404导入本氏烟(Nicotianabenthamiana)。PCR筛选表明,目的片段已整合至转基因植株的基因组DNA中,引起与其同源的基因发生沉默,从而可抑制病毒复制,达到抗该病毒的目的,为防治这类病毒病害提供了新的可能。
The dsRNA in the coat protein (CP) gene of tomato yellow leaf curl China virus (TYLCCNV) was recomposed to plant expression vector pBIN19, and than introduced into Nicotiana benthamiana plants via Agrobacterium tumefaciens mediated transformation. PCR were performed to analyze CP fragment transformation and expression in transgenic plants. The results of PCT indicated that CP gene was successfully integrated into chromosome DNA of transgenic N. benthamiana, and induced its homologous gene silencing to inhibit virus' copy. These results provided a new probability for controlling this kind of virus.
