氢化可的松对新生儿细胞产生IFNγ的影响

摘要 本文观察了氢化可的松对新生儿脐血单个核细胞(CBMC)产生γ-干扰素(IFN_γ)的影响。试检结果表明,药理浓度(10^(-6)M)氢化可的松对ConA 诱导新生儿CBMC产生IFN_γ有明显的抑制作用,其抑制幅度大于该浓度氢化可的松对成人单个核细胞产生IFN_γ的抑制作用。生理浓度(10^(-7)M)氢化可的松对成人单个核细胞生成IFN_γ不产生明显影响,却使新生儿CBMC产生IFN_γ的水平呈下降趋势。将甘草甜素(100μg/ml)与新生儿CBMC预孵育12小时,可部分地逆转药理浓度氢化可的松对新生儿CBMC产生IFN_γ的抑制作用。本文的结果表明新生儿CBMC对氢化可的松抑制IFN_γ产生的效应比成人敏感,提示在儿科临床使用糖皮质激素时,更应警惕其造成的机体免疫功能下降及引起易感的问题。而在使用糖皮质激素前先使用一定量的甘草甜素,有可能会缓解由使用糖皮质激素而引起的免疫抑制效应。 The effect of hydrocortison (HC) on the IFNy production of human neonatal cord blood mononuclear cells (CBMC) was studied. The results revealed that in the presence of pharmacological concentration (10-6M) of HC. the IFNγ production of CBMC induced by ConA was markedly inhibited. The inhibitory effect of HC(10-6M) on IFNγ production was more remarkable in CBMC than that in adult peripheral mononuclear cells. When the concentration of HC was decreased to 10-7M (physiological concentration), the ConA induced IFNγ production of adult cells was not significantly affected, but the level of IFNγ produced by CBMC was lowered, suggesting that CBMC were more sensitive to the inhibitory effect of HC. It also suggested that one must be more cautious to the decline of immune functions and the susceptibility to infections when cortico steroid therapy was used in pediatric cases. However, it was found that the inhibitory effect of HC on IFNγ production could be partially reversed if CBMC were preincubat-ed with g]ycyrrhizin(100ug/ml) for 12hr before ConA stimulation. Administration of glycyrrhizin before corticosteroid therapy may prove to be helpful in minimizing the side effect.
