摘要
研究了FCS,17β-E2,FSH,LH对牛卵泡卵母细胞成熟的作用及培养时间对卵母细胞核和细胞质成熟的影响。结果表明,添加体积分数为10%~15%的FCS,1~2mg·L-117β-E2,20~50mg·L-1LH和10mg·L-1FSH有利于提高卵母细胞核成熟,放出第一极体。而成熟培养20,24h的卵母细胞成熟率显著高于培养16h(P<0.05),核成熟后培养3~4h有利于卵母细胞质成熟,对其后的受精、卵裂及胚胎的进一步发育有重大影响。且TCM199+质量分数10%~15%的FCS+1~2mg·L-117β-E2+10mg·L-1FSH+20~50mg·L-1LH是卵母细胞成熟的理想培养系统。
The effects of FCS,culture time,17β estradial,FSH and LH on the bovine oocytes in vitro maturation were studied.The results suggested that addtions of 10%~15% FCS,1~2 mg·L -1 17β E 2,20~50 mg·L -1 LH and 10 mg·L -1 FSH be effective to improve oocyte nuclear maturation,and to extrude the first polar body.The maturation rate of oocytes cultured for 20,24 h was significantly higher than that of 16h( P<0.05 ).After nuclear maturation,culture for 3~4 h was benefical to oocyte cytoplasm maturation,and had a significant effect on fertilization,cleavage and embryo development.Addition of 10%~15% FCS,10 mg·L -1 FSH 10~50 mg·L -1 LH and 1~2 mg·L -1 17β E 2 in the TCM199 medium were an ideal culture system for bovine oocyte in vitro maturation.
