摘要 目的:建立血浆茶碱浓度监测的高效液相色谱方法。方法:血浆经高氯酸沉淀,以ZORBAXEclipseXDB--C18为色谱柱;流动相为甲醇-水-0.1%TFA体系,流速为0.8mL·min^-1;检测波长为274nm。结果:茶碱浓度在0.25~40.00mg·L^-1范围内线性关系良好(r=0.9999);定量下限为0.25mg·L^-1;高中低三个浓度的相对回收率分别为(98.65±2.24)%、(101.62±1.85)%和(100.92±1.31)%。对54例患者做茶碱血药监测,临床效果满意。结论:本方法准确可靠、简便快速,可用于茶碱血药浓度监测的常规方法。 Objective To develop a high performance liquid chromatography method for the determination of theophlline in plasma. METHODS The plasma were precipitated by perchloric acid. The analytical column was packed with ZORBAX Eclipse XDB--C18. The mobile phase were methanol-- water--0.1% trifluoroacetic acid and the flow rate was 0.8 mL · min^-1. The UV detection wavelength was 274 nm. RESULTS Excellent liner relationship was obtained from the range of 0.25 mg · L^-1 to 40.00 mg · L^-1(r=0. 999 9), the limit determination of doxifluridine was 0.25 mg · L^-1. The relative recoveries were (98.65±2.24) %, (101.62±1.85) %and (100. 92±1.31) % respectively at three concentrations. The clinical application was good. CONCLUSION The method was accurate, simple, rapid and could be used to TDM as a roution method.
