染锰大鼠血清和睾丸LDH活性的变化 被引量:5

摘要 目的研究锰对大鼠血清和睾丸乳酸脱氢酶(lactate dehydrogenase,LDH)的影响。方法将正常雄性SD大鼠48只,随机分为1个对照组和2个染锰组,给予各组大鼠腹腔注射生理盐水和15、30 mg/kg氯化锰。分别染锰4wk和6wk,随机处死各组大鼠8只。应用化学比色法测定血清和睾丸LDH活性。结果①与空白对照组比较,染锰6wk 30mg/kgMnCl2组血清LDH活性显著升高(P<0.01),染锰4wk 30mg/kg MnCl2组,6wk 15mg/kg MnCl2组,6wk30mg/kg MnCl2组睾丸LDH活性均降低(P<0.01)。②染锰时间相同,染锰6w 30mg/kg MnCl2组与15mg/kg MnCl2组比较,血清LDH活性显著升高(P<0.01),染锰4wk和染锰6wk,30mg/kg MnCl2组与15mg/kg MnCl2组相比较,睾丸LDH活性均降低(P<0.05)。③染锰剂量相同,染锰6wk组与染锰4wk组相比较,血清LDH活性显著升高(P<0.01),睾丸LDH活性显著降低(P<0.01)。结论过量锰可诱发大鼠睾丸LDH活性降低,这是生精障碍的重要原因;睾丸LDH活性可作为预测生精障碍的一个客观指标;过量锰诱发大鼠血清LDH活性升高,提示过量锰对心肌、骨骼肌、肾和肝等具有毒性效应。 Objective To study changes of the activity of lactate dehydrogenase(LDH) in serum and testis of rats exposed to Manganese.Methods 48 male rats were randomly divided into 6 groups(blank control 4w,blank control 6w,15 mg/kg MnCl2 4w,15 mg/kg MnCl2 6w,30mg/kg MnCl2 4w,30mg/kg MnCl2 6w),8 rats/group.15mg/kg MnCl2 and 30mg/kg MnCl2 groups were contaminated by manganese(MnCl2.4H2O) by I.P for 4 and 6 weeks respectively and blank control groups was injected with NS by I.P.All rats were injected once a day,five times per week.In the 4th and 6th weekend,their serum and testis were collected to measure by colorimetric analysis the activity of lactate dehydrogenase(LDH) in serum and testis.Results ①Compared with blank control groups,the activity of LDH was increased in serum in 6w 30mg/kg group,and decreased in testis in 4w 30mg/kg MnCl2,6w 15mg/kg MnCl2,6w 30mg/kg MnCl2 manganese exposure groups(P0.01).② Among manganese exposure groups of same time,the activity of LDH was increased more significantly in serum in 30mg/kg group than it in 15mg/kg group among 6w manganese exposure groups(P0.01),but was decreased in testis in 30mg/kg group than it in 15mg/kg group among 4w and 6w manganese exposure groups(P0.05;③Among manganese exposure groups of same dose,the activity of LDH was increased more significantly in serum but decreased in testis in 6w manganese exposure groups than it in 4w manganese exposure groups(P0.01).Conclusions The decreasing activity of LDH in testis might be important cause of spermatogenic disturbance induced by excessive manganese;activity of LDH in testis could be objective indes of prognostic spermatogenic dysfunction.The activity of LDH in serum was increased,It was suggested the excessive manganese can have serivous toxic effect on the heart,skeletal muscle,the liver et al in rats.
