小鼠闭塞性细支气管炎动物模型的建立 被引量:2

摘要 目的通过异位气管移植建立小鼠的闭塞性细支气管炎动物模型,观察其病理改变及免疫因素对模型的影响。方法 C57BL/6小鼠30只作为供体,取出气管分别植入10只C57BL/6小鼠、20只BALB/c小鼠颈部皮下。术后分3组。同质组:10只C57BL/6小鼠。对照组和实验组:每组10只BALB/c小鼠。同质组、对照组小鼠给予生理盐水25mL·kg-1·d-1灌胃。实验组小鼠25mg·kg-1·d-1环孢素A灌胃。术后4周处死小鼠,取出移植气管。观察移植气管的阻塞率、上皮细胞缺失率等指标。结果同质组移植气管管腔通畅,上皮细胞连续完整。对照组移植气管管腔阻塞,气管上皮细胞全部脱落。实验组气管有部分阻塞,部分上皮细胞脱落,气管的阻塞率、上皮细胞缺失率明显低于对照组(P<0.05)。结论小鼠的异位气管移植在术后4周可以观察到与闭塞性细支气管炎相同的病理改变。免疫药物对该病理过程有抑制作用。 Objective To set up an animal model of mice heterotopic tracheal allograft transplantation,and to observe the pathological changes and result contribution by immunity.Methods The trachea was transplanted from C57BL/6 donor mice into a subcutaneous pouch in the dorsal surface of the neck of BALB/c mice and C57BL/6 mice.C57BL/6 mice was isograft group.BALB/c mice were randomly divided into two groups:research group and control group.There were 10 mice in every group.The Isograft group and control group were fed with 0.9%Nacl 25 mg·kg^-1·d^-1.The research group were fed with 25 mg·kg^-1·d^-1 cyclosporine A.After 4 weeks,grafts were taken out and stained by HE.Then the obliteration ratio of allograft and the deletion ratio of epithelium were determined.Results The loss of epithelium and the development of obliterative lesions were observed in the allografts but not the isografts.The obliteration ratio of allograft and the deletion ratio of epithelium of the control group were higher than those of the research group(P〈0.05).Conclusion Animal model of mice heterotopic tracheal allograft transplantation can produce lesions histologically indistinguishable from those observed in obliterative bronchiolitis.Immunosuppressive drug can inhibit development of obliterative lesions in the allografts.
