妊娠糖尿病患者外周血白细胞核因子κB的表达

Expression of NF-κB in peripheral white blood cells in patients with gestational diabetes mellitus

摘要 目的了解妊娠糖尿病(GDM)患者外周血白细胞核因子κB(NF-κB)的表达情况。方法随机入组正常孕妇(NGT组)、妊娠糖尿病孕妇(GDM组)、孕龄期未孕女性(Con组)各10例,以免疫荧光化学染色法和凝胶迁移电泳(EMSA)法检测外周血白细胞NF-κB的表达。结果免疫荧光化学染色结果显示Con组细胞浆染色阳性,细胞核染色阴性;NGT组核内染色轻度阳性;GDM组核内染色显著阳性。EMSA结果显示GDM组、NGT组、Con组灰度相对百分比分别为28.32%、8.66%、5.71%,GDM组与Con组和NGT组间均有统计学差异(P<0.01),但Con组与NGT组间无统计学差异(P>0.05)。结论 GDM患者白细胞核内NF-κB被激活,后者可能参与GDM的发病。 Objective To investigate NF-kB expression in peripheral white blood cells(WBC) in women with gestational diabetes mellitus(GDM), then approach to the relation between morbility of GDM and inflammatory reaction. Methods We collected the anticoagulated blood from the subjects of control, NGT and GDM groups, then separated leukocyte for detecting the activity of NF-kB, for immunofluorescent staining of smeared cells and for electrophoretic mobility shift assay (EMSA) after abstracting nucleoprotein. Results Immunofluorescent staining showed that cytosol was positive and nuclei was negative of peripheral blood leucocyte in control group, while nuclei were slightly positive in NGT group and obviously positive in GDM group. EMSA-indicated gray scale (percentage) was 28.32% in the GDM group,5.71% in NGT group and 8.66% in control group. The gray scal level was much higher in GDM than in NGT and control group(P〈0.01), with no significant difference between NGT group and control group(P〉0. 05). Conclusions NF-kB is significantly activated in GDM group, which may participate in the patbogenesis of GDM.
