摘要
利用ISSR分子标记研究了辽宁9种耐寒杜鹃花的亲缘关系,从100个引物中筛选出13个能扩增出清晰带型并具多态性的引物,共扩增出了576条DNA片段,分子量为300~2000 bp,平均每条引物扩增出44.31条DNA片段。采用DPS软件,计算9种杜鹃花的Nei氏距离,并用UPGMA法构建了系统树。结果表明,9种杜鹃花明显聚为3大类:照白杜鹃、牛皮杜鹃聚为一类;迎红杜鹃、缘毛迎红杜鹃、兴安杜鹃聚为一类;黄杨杜鹃、淀川杜鹃、红枫杜鹃和大字杜鹃聚为一类,其中第2类与形态学分类结果一致。
Genetic relationship of nine species in Rhododendron was studied using inter-simple sequence repeat(ISSR) marker.Thirteen primers were screened out from 100 random primers and a total of 576 DNA fragments were amplified ranging from 300-2 000 bp.The average number of DNA fragments produced by each primer was 44.31.Nei'S genetic distances were calculated by using DPS software and the dendrogram were constructed by using UPGMA.The nine Rhododendron species were divided into three groups: R.micranthum and R.aureum were clustered into one group;R.mucronulatum f.ciliatum,R.dauricum and R.mucronulatum were clustered into the second group,which was in consistence with the morphological classification.R.schlippenbachii,R.hongfeng,R.yedoehse huangyang,and R.yedoehse were clustered into the third group.
