摘要
目的为配合3,4,5-三羟基苯甲酸(trihydroxybenzoic acid,TBA)脂质体的制备工艺研究,建立测定TBA脂质体中药物含量及包封率的高效液相色谱(HPLC)方法。方法超声薄膜法制备TBA脂质体,HPLC测定药物含量及包封率。色谱柱为AgilentTC-C18柱(4.6mm×150mm,5μm),流动相为甲醇-0.1%磷酸(15:85),流速为0.8mL.min-1,检测波长为273nm。结果 TBA在50~150mg.L-1内线性关系良好(r=0.9980,n=5),平均回收率为98.25%(RSD=0.45%)。结论超声薄膜法可以制备出包封率较高的TBA脂质体,HPLC测定脂质体的包封率,方法准确可靠,简单快速。
OBJECTIVE To screen the optimal formulation and preparation of trihydroxybenzoic acid (TBA) liposomes,and establish a HPLC method for the determination of the content and entrapment efficiency of TBA liposomes.METHODS TBA liposomes were prepared by the ultrasonic membrance method.The content of TBA and the entrapment efficiency were detected by HPLC.The analytical column was an Agilent TC-C18 column(4.6 mm×150 mm,5 μm).The mobile phase was methanol-0.1% phosphoric acid(15:85).The flow rate was 0.8 mL·min-1.The detection wavelength was 273 nm.RESULTS TBA showed good linerity over the range fo 50-150 mg·L-1 (r=0.998 0,n=5).The average recovery was 98.25%(RSD=0.45%).CONCLUSION The entrapment efficiency of TBA liposomes which was prepared by this method is up to the requirement.HPLC was proved to be simple,accurate,sensitive and applicable.
