CCND1基因不同转录本在宫颈癌组织中的表达及意义 被引量:3

摘要 目的探讨CCND1基因长短两种转录本及其蛋白产物cyclinD1在宫颈癌组织中的表达情况及对宫颈癌生物学行为的影响。方法采用实时定量RT-PCR和免疫组化S-P法检测CCND1基因两种转录本及CyclinD1蛋白在宫颈癌组织和正常组织中的表达。结果 CCND1基因的短转录本在正常组织和宫颈癌组织中相对表达量有统计学意义(P<0.05)而长转录本则无统计学意义(P>0.05);并且在宫颈癌组织中发现短转录本转录量在低分化组织中降低明显,有统计学意义(P<0.05);同时发现cyclinD1蛋白在宫颈癌组织中部分高表达(57.14%),在正常宫颈组织中仅在具有分化潜能的基底细胞中有所表达(13.33%)。结论 CCND1基因的短转录本对于宫颈癌可能存在保护作用,同时,CCND1基因的两种转录本的相对表达量的差异不会影响cyclinD1蛋白总的表达量。 Objective To investigate the expressions of 2 alternative transcripts of CCND1 gene and its protein product cyclin D1 in cervical cancer and the effect of their expressions on the biological behavior of cervical cancer.Methods We detected the expressions of these 2 transcripts and cyclin D1 in cervical cancer tissue and normal tissue using real-time reverse transcription polymerase chain reaction and immunohistochemistry.Results The expression of short transcript in cancer tissue was significantly lower than that in normal tissues(P = 0.001).No significant difference in the expression of long transcript was found between cancer and normal tissues(P =0.182).In cancer tissue,the expression of short transcript was significantly lower in poorly differentiated tissue than in highly differentiated tissue(P =0.001).Cyclin D1 was highly expressed in part of the cancer tissue,and it was only expressed in the basal cells with differentiation potential in the normal tissue.Conclusion The short transcript of CCND1 gene may have a protective effect in cervical cancer,and the relative expression level of 2 transcripts of CCND1 gene may not affect the expression of cyclin D1.
