黄粉虫抗菌肽对K562细胞周期的影响及与羟基脲对细胞增殖抑制的比较 被引量:2

摘要 目的观察黄粉虫抗菌肽对K562细胞周期的影响;比较该抗菌肽与羟基脲对K562细胞的增殖抑制作用。方法用流式细胞仪检测黄粉虫抗菌肽对K562细胞周期的影响。黄粉虫抗菌肽与羟基脲分别设置5个浓度组,用MTT比色法分别求出2种药物不同浓度对K562的抑制率,根据浓度和抑制率用SPSS软件求出回归方程和IC50。结果细胞周期G0/G1期和G2/M期细胞比例增加,S期细胞比例下降。其中只有G0/G1期和S期与对照组相比,差异具有统计学意义(P<0.01)。说明抗菌肽可使细胞周期阻滞于G0/G1期,从而抑制S期DNA的合成。抗菌肽和羟基脲的IC50分别为29.98和3644.45μg/ml。结论抗菌肽可通过影响细胞周期来发挥抗K562细胞的作用。抗菌肽明显优于羟基脲对K562细胞的增殖抑制作用,有较好的应用前景。 Objective To assess the effects of antimicrobial peptides of Tenebrio molitor Linnaeus on K562 cells,and to compare the inhibitory effects of these peptides and of hydroxyurea on K562 cell proliferation.Methods A flow cytometry was employed to identify the anti-K562 effects of molitor antimicrobial peptides.Five parallel concentrations of the two agents were set up for comparison,and their K562 inhibition rates at different concentrations were determined using the MTT colorimetric assay.The regression equation and IC50 were computed using the SPSS software based on the relations between the concentration and inhibition rates.Results The proportion of cells in G0/G1 and G2/M phase increased,and that of cells in S phase decreased.Statistical differences were only noticed by comparing cells in G0/G1 and S phases and the control group(P0.01),indicating that antibacterial peptides inhibited DNA synthesis in S phase by G0/G1 phase arrest.The IC50 of antibacterial peptides and hydroxyurea were 29.98 and 3644.45 μg/ml,respectively.Conclusion The anti-K562 activity of antibacterial peptides,a result of cell cycle impairment,noticeably outstripped the inhibitory effects of hydroxyurea on K562 cell proliferation.These peptides have a good prospect for practical application in future.
