兔出血症病毒免疫早期AF细胞和AS细胞的动态 被引量:1

KINETICS OF AFC AND ASC AT THE INITIAL STAGE AFTER IMMUNIZATION OF RHDV

摘要 本试验采用间接免疫荧光法和单层液相凝集新技术,分别检测 RHDV(RabbitHemorrhagic Disease Virus)免疫早期特异性抗体形成细胞(Antibody-Forming Cell-AFC)和抗体分泌细胞(Antibody-Secreting Cell—ASC)在各免疫淋巴器官中的发生和动态变化。结果,RHDV 免疫后24小时就可在脾脏中检测到 AFC,48小时出现 ASC,以后两类细胞不断增加,到120小时 AFC 呈继续增长趋势,而 ASC 则趋向稳定。在淋巴结,48小时开始出现 AFC,72小时检出 ASC,到120小时两类细胞继续增长。在胸腺组织中,分别于24小时和48小时开始检测到低水平的 AFC 和 ASC,随免疫期延长,无明显增长趋势。盲肠末端在72小时和120小时测到极少量的 AFC,而直至120小时仍未查到 ASC。血液淋巴细胞到120小时两类细胞均未检测到。同期测定血清 HI 抗体,到168小时才有低效价的 HI 抗体出现。 Indirect immunofluorescent assay and monolayer liquid phase hema- gglutination assay were used respectively to detect Antibody-Forming Cell (AFC) and Antibody-Secreting Cell (ASC) specific to Rabbit Hemorrhagic Di- sease Virus (RHDV) at the initial stageof immunization of RHDV.AFCs were first detected hours 24 post-immunization (PI) in spleen and hours 48PI in lymph node,and then rose continuedly until hours 120PI in both organs.ASCs appeared first at hours 48PI in spleen and hours 72 PIin lymph node,and then their number rapidly increased.At hours 120 PI,ASCs tended to maintain a stable status in spleen.In thymus tissue,lower levels of both AFCs and ASCs cold be detected respectively from hours 24 and 48 PI to hours 120 PI and no increase occurred.In appendix,only a very limited number of AFCswere measured at hours 72 and 120PI and no ASCswere seen from hours 24 to 120 PI.No both AFCs and ASCs were detected from hours 24 to 120PI among the blood lymph- ocytes.Alower-titre HI antibodies to RHDV were measured at hours 168 PI in sera.
