外加稳恒直流电场对兔球囊损伤腹主动脉舒张功能的影响

Effect of the applied external steady direct current electrical fields on diastolic function after balloon injury in the rabbit abdominal aorta

摘要 目的:观察外加稳恒直流电场对兔球囊损伤后腹主动脉舒张功能的影响。方法:健康日本大耳白兔40只,随机分为4组,建立兔腹主动脉球囊损伤模型,在腹主动脉两侧腰大肌埋置刺激电极。电场组术后给予稳恒直流电场刺激(电场强度4V/cm,30min/d),对照组术后不给予电场刺激,电场加N硝基-L-精氨酸甲酯(L-NAME)组和电场加L-精氨酸(L-Arg)组在术后电场刺激基础上分别经腹腔注射给予L-NAME及经胃管给予L-Arg治疗,术后1、2、4周检测血管舒张功能,硝酸还原酶法检测血管组织匀浆一氧化氮(NO)。结果:术后4周电场组对乙酰胆碱的最大舒张反应强度及NO水平[(64.65±0.53)%、(652.18±2.66)μmol/L]均显著高于对照组[(51.97±0.23)%、(637.88±5.68)μmol/L]及电场加L-NAME组[(49.57±0.82)%、(628.29±4.07)μmol/L],差异有统计学意义P<0.05,但显著低于电场加L-Arg组[(71.29±0.82)%、(657.14±3.88)μmol/L],差异有统计学意义(P<0.05)。结论:兔腹主动脉球囊损伤后外加稳恒直流电场干预4周后血管内皮舒张功能较对照组显著改善,提示外加电场有利于血管内皮功能的恢复。上述作用可能是通过NOS-NO信号通路改善血管内皮舒张功能的结果。 Objective:To observe the effects of the applied external steady direct current electrical fields on diastolic function after balloon injury in the rabbit abdominal aorta.Method:The model of balloon injury abdominal aorta of rabbit was established and the platinum wire electrode was implanted in the psoas major muscle in the two sides of abdominal aorta of rabbit.40 healthy Japanese white rabbits were randomly divided into 4 groups.The rabbits of electrical fields group were given electric stimulation after balloon injury (electric field strength:4 V/cm,30 minutes/day),But the rabbits of control group were not.The rabbits of electrical field+L-NAME group and electrical field+L-Arg group were given L-NAME by intraperitoneal injection and L-Arg by gastric tube respectively on the basis of electric stimulation after balloon injury.The rabbits were sacrificed on 1st,2nd,4th week after balloon injury.Vascular diastolic function was observed and the level of vascular tissue NO was detected though nitrate reductase.Result:The maximum relaxation response intensity to Ach and the level of NO of the rabbits in electrical field group [(64.65±0.53)%,(652.18±2.66)μmol/L] was significantly higher than that of the rabbits in control group [(51.97±0.23)%,(637.88±5.68)μmol/L] and electrical field+L-NAME group [(49.57±0.82)%,(628.29±4.07)μmol/L] (P〈0.05) and lower than that of the rabbits in electrical field+L-Arg group [(71.29±0.82)%,(657.14±3.88)μmol/L] (P〈0.05) at 4th week after balloon injury.Conclusion:The vascular endothelial function of the rabbit abdominal aortal after ballon injury,which was stimulated by the external steady direct current electrical field 4 weeks,compared with that of the control group had significantly improved.It indicated that the external electrical field was favorable for the recovery of endothelial functions.The improvement of vascular endothelial function maybe come true though the NOS-NO signaling pathway.
