摘要
目的:观察牛磺酸(Tau)预处理对大鼠肢体缺血再灌注(IR)后内质网应激反应的影响。方法:制作大鼠肢体IR损伤动物模型,将大鼠随机分成对照组、IR组和Tau组,测定各组骨骼肌组织湿/干(W/D)、活性氧(ROS)、丙二醛(MDA)、黄嘌呤氧化酶(XOD)、超氧化物歧化酶(SOD)含量,采用Western blot和实时聚合酶链反应法测定骨骼肌内质网应激(ERS)蛋白-葡萄糖调节蛋白94(GRP94)表达水平,分析牛磺酸对肢体IR损伤与ERS的关系。结果:与IR组比较,Tau组大鼠骨骼肌组织ROS、MDA、XOD和W/D均显著降低(P<0.05,P<0.01),而SOD活性显著增加(P<0.05),GRP94在蛋白及mRNA水平表达下调。结论:牛磺酸对肢体IR损伤的保护作用与减轻骨骼肌ERS反应有关。
Objective:To investigate the effect of taurine on endoplasmic reticulum stress after limbs ischemia-reperfusion (IR)in rats.Methods:Making the model of limbs ischemia reperfusion injury in rats.Rats were randomly divided into 3 groups(n=10):control group,IR group and taurine(Tau)group.The contents of reactive oxygen species(ROS),malondialdehyde(MDA),xanthine oxidase(XOD),superoxide dismutase(SOD) and rario of wet/dry (W/D)of skeletal muscle in different group rats were detected.Then Western blot and real time-polymerase chain reaction were used to examine the expression change of glucose regulated protein(94GRP94) of skeletal muscle in different group rats.Results:Compared with control group,the content of ROS,MDA,XOD and W/D in skeletal muscle increased significantly in IR group.The level of SOD in skeletal muscle decreased after reperfusion.The expression of GRP94 protein and GRP94 mRNA increased significantly in IR group.And taurine can down-regulate GRP94 expression in skeletal muscle.Conclusion:The protective role of taurine on limbs IR is related to decrease endoplasmic reticulum stress in rat skeletal muscle.
