针刀松解法或电针对膝骨关节炎兔脑组织cAMP、cGMP的影响 被引量:7

Effects of acupotomy or electroacupuncture on brain cAMP and cGMP in rabbits with knee osteoarthritis

摘要 目的对比观察针刀松解法和电针对膝骨关节炎兔脑组织中3’,5’-环磷酸腺苷(cAMP)、3’,5’-环磷酸鸟苷(cGMP)含量的影响。方法将52只新西兰兔随机分为正常组、骨关节炎模型组、针刀治疗组和电针治疗组。伸直位制动兔膝关节6周,制备兔膝骨关节炎模型,针刀松解法或电针治疗3周,治疗结束1周后,采用Lequesne MG的关节局部反应和关节畸形评估级别对兔膝关节进行评价,应用放射免疫法比较脑组织cAMP、cGMP含量的变化。结果关节局部反应和关节畸形率,模型组Ⅱ’级占的百分率较正常组明显升高,针刀或电针治疗后Ⅰ’级占的百分率较模型组明显升高;下丘脑和海马部位模型组cAMP含量明显升高,针刀治疗后,丘脑和海马部位cAMP明显降低;电针治疗后,海马部位cAMP含量明显降低,丘脑部位cAMP含量无明显变化。下丘脑和海马部位模型组cGMP含量较正常组虽有下降趋势,但无显著差异;针刀或电针治疗对下丘脑和海马部位的cGMP亦无明显影响。结论针刀松解法或电针对关节局部反应和关节畸形有一定的改善作用;针刀松解法和电针均能阻抑海马部位cAMP的生成,且针刀松解法尚可阻抑丘脑部位cAMP的生成;而针刀松解法或电针对丘脑部位和海马部位的cGMP含量无明显影响。提示两种疗法能减轻骨性关节炎疼痛、阻抑关节软骨损伤和功能改变等,可能与针刀松解法或电针能够调节下丘脑部位和海马部位细胞内第二信使cAMP水平相关。 Objective To compare and observe the effects of acupotomy or electroacupuncture on the content of brain cAMP and cGMP in rabbits with knee osteoarthritis(KOA) .Methods New Zealand rabbits(n=52) were randomly divided into the normal group,model group,acupotomy group and electroacupuncture group.The rabbit model of KOA was established by fixing rabbit knee straightly for 6 weeks,and then the rabbits were treated with acupotomy or electroacupuncture for 3 weeks.After the treatment finished for one week,the rabbit knee was reviewed by using the standard of articular local reaction and joint deformity of Lequesne MG method.The content changes of brain cAMP and cGMP were detected by applying radioimmunoassay.Results The percentage ofⅡ'rank of local joint reaction and joint deformity increased significantly in the model group compared with the normal group.The percentage ofⅠ'rank increased significantly in the acupotomy group and electroacupuncture group compared with the model group.After acupotomy treatment the content of cAMP in the hypothalamus andhippocampus decreased significantly,and after electroacupuncture treatment the content of cAMP decreased significantly in the hippocampus but did not changed in the hypothalamus.Although the content of cGMP in the hypothalamus and hippocampus tended to decrease in the model group compared with the normal group,there was no significant difference.The treatment of acupotomy or electroacupuncture had no obvious effects on the content of cGMP in the hypothalamus and hippocampus. Conclusion Acupotomy and electroacupuncture can relieve the articular local reactions and joint deformity,and inhibit the formation of cAMP in the hippocampus.Acupotomy can inhibit the formation of cAMP in the hypothalamus,while both therapies have no obvious effects on the content of cGMP in the hypothalamus and hippocampus.The results indicates that the two therapies can relieve the pain of KOA,and inhibit the injury of articular cartilage and functional changes,which may be related to the regulation of the two therapies on intracellular second messenger-cAMP in the hypothalamus and hippocampus.
