卡托普利对大剂量维生素D_3联合动物油诱导大鼠心血管系统钙化的预防作用 被引量:1

摘要 目的观察大剂量维生素D_3联合动物油诱导大鼠心血管系统钙化的病理改变,以及卡托普利对该病变的预防作用。方法选择6月龄SPF级雄性SD大鼠24只,随机分为对照组、模型组、卡托普利组,每组8只。制作主动脉弓和心脏组织病理切片观察钙化病变,胸主动脉大体Von Kossa染色后计算钙化面积百分比,同时检测血脂和血清钙、磷水平。结果与对照组比较,模型组大鼠胸主动脉钙化受损总面积明显增大,HDL-C明显降低,血清磷水平明显升高(P<0.05);与模型组比较,卡托普利组大鼠胸主动脉受损总面积比较无显著差异,HDL-C水平恢复至对照组水平。结论大剂量维生素D_3与动物油诱导可致大鼠心血管系统出现类似于动脉粥样硬化性钙化,卡托普利对钙化病变有预防作用。 Objective To observe the pathological change of calcified vascular system and myocardium induced by vitaminD3 and animal oil and the preventive action of captopril which is angiotensinconverting enzyme inhibitor.Methods Twenty-four SD rats of six months old were randomly divided into three groups including control group,model group and captopril therapy group with eight rats in each group.Six weeks later,all animals were sacrificed,the aortic arch,ventricle of heart and thoracic aorta were taken to make 4 micrometer sections which were stained with hematoxylin and eosin.The pathological changes were observed under light microscope,and the percentage of calcification area of thoracic aorta stained by argent nitrate was calculated using imagepro plus 6.0 software and the content of calcium,phosphate,high density lipoprotein cholesterol,low density lipoprotein cholesterol,triglyceride and total cholesterol in blood serum were detected.Results Compared with the control group,model group had significant vascular calcification,vascular smooth muscle cells were transformed into cartilagelike cells,percentage of calcified area of thoracic aorta and serum phosphate level increased significantly,while serum high density lipoprotein cholesterol decreased significantly(P〈0.05).Compared with model group,the damaged area of thoracic aorta had no significant difference in captopril therapy group,the content of high density lipoprotein cholesterol in serum recovered to the level of control group.Conclusion Large dose of vitaminD3 plus animal oil can induce significant calcification of cardiovascular system similar to atherosclerotic calcification.Captopril has preventive effect against the calcification of cardiovascular system,and can increase the content of high density lipoprotein cholesterol in serum.
