摘要
以生产上农艺性状表现良好的5个低酚陆地棉品种为材料对体细胞胚胎发生和植株再生进行了研究,供试5个品种均得到了再生植株,初步建立了低酚棉再生体系。研究结果表明,以无菌苗下胚轴为外植体,在附加IAA,KT,2,4-D各01mg/L的MS无机元素+B5有机成分的培养基上,易诱导出灰软疏松的愈伤组织,当去掉2,4-D,且NH4NO3减半后,胚性愈伤组织大量出现,再除去激素,KNO3加倍有利于胚状体的发生。在不含激素但含活性炭15g/L的SH培养基上,子叶期胚状体可以萌发,并长出具有根、茎、叶的完整植株。低酚棉再生体系的建立为棉花的基因转化奠定了基础。
