雷公藤多苷对大鼠佐剂性关节炎的治疗作用与NO的关系

The relationship between anti arthritis effect of tripterygium wilfordii polyglycosidium(TWP) and the effect of TWP on NO level

目的探讨雷公藤多苷对大鼠佐剂性关节炎（ＡＡ）的治疗作用与ＮＯ的关系。方法采用镉还原柱层析和Ｇｒｉｅｓｓ比色法，测定ＡＡ模型大鼠血中ＮＯ－２／ＮＯ－３的水平，并结合病理组织学检查和ＮＡＤＰＨ－α组化观察关节处的变化。于致炎后ｄ１９起，每日给予雷公藤多苷２、８和３２ｍｇ·ｋｇ－１。结果雷公藤多苷８和３２ｍｇ·ｋｇ－１组左、右两侧足跖的继发性肿胀度均于给药后ｄ４即明显较给药前小。而且给药后ｄ８，这两组在减轻足跖肿胀的同时，均可显著降低血清中ＮＯ含量。踝关节病理显示：模型对照组关节滑膜组织内有大量炎性细胞（主要是淋巴细胞、巨噬细胞）浸润，关节面不完整，关节腔内可见退化、脱落的滑膜组织碎片，ＮＡＤＰＨ－α组化显示：膝关节滑膜下结缔组织中有散在的蓝黑色ＮＯＳ阳性细胞；而给予雷公藤多苷８和３２ｍｇ·ｋｇ－１后，关节病理观察结果显示：滑膜组织内淋巴细胞、巨噬细胞明显减少，且３２ｍｇ·ｋｇ－１组作用更明显，滑膜下结缔组织中ＮＯＳ阳性细胞消失。结论在大鼠佐剂性关节炎病理过程中有ＮＯ参与。

AIM To study the relationship between anti arthritis effect of TWP and its effect on NO level in serum of adjuvant arthritis(AA) rats. METHODS NO - 2/NO - 3 stable end products of NO, levels in serum which were detected with cadmium reduction column and griess colorimetry. Tissue sections of the ankle joint were stained with HE method, and those of the synovisal tissues in knee joint with NADPH α histochemistry. RESULTS 2, 8, 32 mg·kg -1 of TWP were given via ig from the 19th day after infected with Freunds complete adjuvant. 8 and 32 mg·kg -1 of TWP could inhibit the edema degrees of the inflamed paws, and reduce NO level in serum ( P <0 05 and P<0 01 ). HE staining showed that edema degree of inflamed ankle joint was reduced in severity, and inflamed cells (including lymphocytes and macrophages) in synovisal tissues were reduced after 8 and 32 mg·kg -1 of TWP were given; NOS positive cells in synovisal tissue in AA rats disappeared after TWP was given. CONCLUSIOS TWP reduces NO level in serum in rats with adjuvant arthritis, which suggests that of the anti AA actions of TWP is due to its inhibition of NO level.
