摘要
以BSA(牛血清白蛋白)和钙基蒙脱石为原料,研究了BSA在蒙脱石表面上等温吸附、不同pH下的吸附和解吸、不同偶联剂处理的吸附和解吸,并对吸附后上清液和解吸液进行紫外光谱分析。结果表明低pH和偶联剂KH570的使用有利于蛋白质在蒙脱石片层上的吸附,偶联剂存在则更有利于蛋白质的解吸。解吸液中的BSA的共轭效应减弱,上清液中的蛋白质的共轭效应更强。但偶联剂处理后蛋白质结构变化较小。此项研究为药物负载等方面研究奠定基础。
BSA and Ca-montmorillonite as the raw materials,the adsorption isotherm, the adsorption and desorption behavior under different pH values and the usage of coupling agents KH570 or GA were studied. Then by ultraviolet scane the structure change of BSA in supernatant and desorption solution was analyzed. The results showed that low pH and the use of KH570 are conducive to protein adsorption and the coupling agents are helpful for desorption. Analysis of the UV/VIS spectra finds that the conjugated effect of protein released from the interlayer of Montmorillonite weakened while protein in supernatant solution intensified. But the structure of BSA adsorption and desorption after modified by coupling agents changes less in degree. The study lay a foundation for research of drugs loading and so on.
