摘要
目的:检测位于人染色体3p14.2区的FHIT基因在肺鳞癌中的缺失率,探讨FHIT基因在国人肺鳞癌中的作用。方法:收集手术切除21例肺鳞癌及对应正常肺组织标本,应用RT-PCR及DNA测序技术检测FHIT基因的缺失情况。结果:21例正常肺组织中均有FHIT基因的完整表达,10例(47.6%)肺鳞癌标本中检测出FHIT基因mRNA转录本的异常表达,经测序证实为由于FHIT基因多个外显子缺失所致。结论:首次在国人中证实,位于3p14.2的FHIT基因在肺鳞癌中存在较高的缺失率,提示FHIT基因为候选抑癌基因,在肺鳞癌的发生发展中起重要作用。
Objective: To detect the deletion rate and to study the role of the fragile histidine triad (FHIT) gene at 3p14.2 in Chinese squamous cell carcinoma (SQC) of the lung.Methods: Matched normal and cancerous tissues from 21 patients with primary pulmonary SQC were obtained immediately after surgery. Total RNA was extracted, the FHIT gene was detected by RT PCR and DNA sequencing technology.Results: Normal sized FHIT transcript was detected in all of 21 cases of normal matched tissues. Aberrant transcripts were observed in 10(47.6%) of 21 cases with SQC of the lung and the sequence analyses of the aberrant cDNAs revealed absence of various regions between exons 4 and 10.Conclusion: High deletion rate of the FHIT gene in Chinese SQC of the lung suggested the FHIT gene, a candidate tumor suppressor gene at 3p14.2, could be important during tumorigenesis of lung carcinomas.
