摘要
目的探讨通心络超微粉对心肌梗死大鼠转化生长因子-β1(TGF-β1)的表达作用。方法 130只SD大鼠随机选10只为假手术组(0.9%NaCl灌胃4w,n=10,至实验终点时剩有8只)4w后处死。120只SD大鼠结扎冠状动脉前降支,术后存活的38只大鼠随机分为:NaCl灌胃组(0.9%NaCl灌胃4w,n=9),通心络超微粉治疗组(1.0g.kg-1.d-1)10只、卡托普利治疗组(15mg.kg-1.d-1,n=10)10只,药物灌胃4w。用逆转录-聚合酶链反应法(RT-PCR)及免疫组化方法检测TGF-β1的mRNA和蛋白表达情况。结果与假手术组相比,心肌梗死模型组TGF-β1的mRNA和蛋白表达均明显升高(均P<0.01)。与心肌梗死模型组相比,通心络超微粉治疗组和卡托普利治疗组TGF-β1的蛋白和mRNA表达均明显减低(P<0.01)。结论大鼠心肌梗死后TGF-β1表达增加,提示TGF-β1在梗死后心室重构起重要作用。通心络超微粉抑制大鼠心肌梗死后TGF-β1表达基于通心络超微粉的抗氧化应激作用。通心络超微粉对心肌梗死后心室重构的保护作用是有益的。
Objective To investigate inhibitory effects of Tongxinluo on TGF-β1 expression of myocardial infarction rats.Methods 60 rats were randomly divided into sham group (the rats were fed with 0.9% NaCl for 4 weeks,n=10,after 4 weeks the number of rats was 8) and acute myocardial infarction (AMI) group.50 AMI rats underwent the left descending coronary artery ligation and then 28 surviving rats were randomly assigned to AMI model group (the rats were fed with 0.9% NaCl for 4 weeks,n=9),high dose Tongxinluo group (the rats were fed with 1.0 g·kg-1·d-1,n=10),and low dose Tongxinluo group (the rats were fed with 0.05 g·kg-1·d-1,n=9).All the groups were fed with the medicine for 4 weeks.At the end of the experiment all rats were weighted.The heart of rat was fixed and pathologically analyzed.Myocardial infarction areas were quantified by TCC.Morphological characteristics were evaluated with HE staining.Toluidine blue staining showed mast cells in myocardial tissues of rats,the mRNA expression of TGF-β1 was examined by RT-PCR.Results Infarction areas of heart in rats had no obvious difference among every group.Compared to sham operation rats,muscle fibers and the number of mast cell in heart of model group obviously increased,while those of high dose Tongxinluo group decreased and low dose Tongxinluo group had no significant difference.The expression of TGF-β1 mRNA was increased in model group (P0.01).Compared with model group,the TGF-β1 expression was significantly lower in high dose Tongxinluo group and there was no changes in low dose Tongxinluo group.Conclusions TGF-β1 plays key role in ventricular remodeling and myocardial fibrosis.The TGF-β1 participates in the early period of ventricular remodeling and myocardial fibrosis.Ventricular remodeling is closely correlated with myocardial fibrosis.Preventing and treating ventricular remodeling and myocardial fibrosis with Tongxinluo is effective.
