含溶栓颗粒血清对肿瘤坏死因子-α损伤大鼠脑微血管内皮细胞间粘附分子-1的影响

摘要 目的探讨溶栓颗粒对肿瘤坏死因子-α(TNF-α)损伤血管内皮细胞间粘附分子-1(ICAM-1)表达的影响。方法用TNF-α(40ng/mL)损伤大鼠脑皮质微血管内皮细胞(BMECs)造模,用含大、中、小剂量溶栓颗粒和中等剂量阿托伐他汀的大鼠血清干预受损BMECs,应用HE染色观察细胞形态变化,免疫组化法观察ICAM-1的表达。结果 TNF-α可以造成BMECs损伤,表现为细胞数量下降、皱缩。受损细胞的ICAM-1表达明显增多。含大、中、小剂量溶栓颗粒和阿托伐他汀的大鼠血清能减少ICAM-1表达。中等剂量阿托伐他汀的这一作用与大、中剂量溶栓颗粒相当。结论溶栓颗粒可以减少因炎性介质TNF-α增加而受损的血管内皮细胞ICAM-1表达,或许可以减缓动脉粥样硬化形成,并对稳定动脉粥样斑块有积极作用。 Objective To study the effect of Rongshuan Grain on the expression of ICAM-1 of brainmicrovascular endothelial cells (BMECs) injured by TNF-α. Methods TNF-α (40 ng/mL) was used to damagethe BMECs. The serum of rats containing different dosage of Rongshuan Grain and moderate dosage ofAtorvastatin were used to intervene the damaged BMECs. The morphological changes was observed by HEstaining and the expression of ICAM-1 by immunohistochemical method. Result TNF-α damaged theBMECs, manifestations included that the cell number decreased, the cell shape changed to crimple. Theexpression of ICAM-1 of damaged BMECs increased. The serum containing high, moderate and small doseof Rongshuan Grain and moderate dose of Atorvastatin reduced the expression of ICAM-1. The effect ofmoderate dose of Atorvastatin was the same as high and moderate dose of Rongshuan Grain. ConclusionRongshuan Grain can reduce the expression of ICAM-1 of BMECs damaged by TNF-α. The function maydecrease the formation of atherosclerosis and be effective on the stabilization of atheromatous plaque.
