大孔吸附树脂分离纯化青番茄中的番茄苷 被引量:4

摘要 目的研究探讨大孔吸附树脂分离纯化青番茄中番茄苷的工艺。方法采用HPLC-MS/MS检测青番茄中的番茄苷,以番茄苷浓度为指标,考察大孔吸附树脂分离纯化番茄苷的工艺。结果 HPD-100A大孔吸附树脂最佳工艺条件为:柱体积510mL,上样体积为6BV,先用蒸馏水洗脱,再用95%乙醇洗脱,洗脱体积共为6BV。番茄苷粗品经上述工艺纯化后,含量达到26.48%。结论 HPD-100A大孔吸附树脂对番茄苷有较好的吸附分离性能,是分离纯化番茄苷的最佳大孔吸附树脂。 Objective To improve the content of tomatine in green tomatoes by separation and purificationof tomatine with macroporous adsorption resin. Method HPLC-MS/MS methods were established todetermine the tomatine in green tomatoes. With the concentration of tomatine as the main index, thetechnologies of separation and purification of tomatine with macroporous adsorption resin were studied.Result The optimal technologies of HPD-100A macroporous absorption resin were as follows:pillar volumewas 510 mL, the volume of sample added was 6 BV. The column was washed by distilled water and then95% ethanol, the elution volume was 6 BV. The content of tomatine was 26.48% in crude tomatine afterpurification with above technologies. Conclusion The adsorption and separation capability of HPD-100Amacroporous absorption resin was good, and it was the optimal resin.
