苯酚-硫酸法测定青钱柳中多糖含量 被引量:37

Determination of Polysaccharides from Cyclocarya paliurus(Batal.)Iljinskja by Phenol-sulfuric Acid

摘要 建立一种苯酚-硫酸分光光度法测定青钱柳多糖的方法。采用先除杂后水提分离提取青钱柳多糖,以葡萄糖标准液绘制标准曲线,用苯酚-硫酸法比色测定青钱柳多糖含量,检测波长488 nm。并研究了显色时间、显色温度、苯酚及硫酸用量对糖含量测定的影响。结果显示:加入1 mL 5%的苯酚溶液,硫酸5 mL,显色温度100℃,在水浴中加热20 min时测定条件最优。该方法快速、显色稳定、重现性好、平均回收率98.2%,RSD2.5%,适用于青钱柳多糖的含量测定,测得3批青钱柳多糖的平均含量为4.47%。 A simple phenol-sulfuric acid spectrophotometry method was developed to determine the contents of polysaccharides from Cyclocarya paliurus (Batal.) Iljinskja. The polysaccharides in Cyclocarya paliurus (Batal.) Iljinskja samples were purified by precipitation with ethanol followed by extracted with distilled water. The contents of the polysaccharides were determined by phenol-sulfuric acid colorimetrictitration method at 488 nm with glucose as standard. The influence of different time, temperature, different contents of phenol and sulfuric acid on the contents test of polysaccharide was also investigated. Optimum operating conditions were found to be 5% phenol 1 mL, sulfluric acid 4 mL, temperature 100℃ and time 20 min. At this optimum point, the results obtained showed that the method used in this paper was simple, available, stable and well reproducible, and the average value of the recovery for the determination of the polysaccharides was 98.2% with 2.5% of RSD. The average content of polysaccharide in 3 batches of Cyclocarya paliurus (Batal.) Iljinskja substance was 4.47%. It indicated that the phenol-sulfuric acid method is suitable for the determination ofpolysaccharide from Cyclocarya paliurus (Batal.) Iljinskjask.
