流式细胞术诊断疟疾的初步应用

Application of flow cytometry as a diagnostic test for malaria

目的利用流式细胞术检测疟原虫感染患者的血液红细胞,建立疟疾诊断的辅助方法。方法采集镜检确诊的疟疾患者血样,同时设正常人血样为阴性对照,通过DRAQ5单染色,利用流式细胞仪进行检测。结果正常人全血、加有DRAQ5荧光染料的正常人全血、未加DRAQ5染料的疟疾患者血样的SSC/FL3-H散点图中未发现有染料标记的红细胞;而在疟原虫感染的患者血样中,有染料DRAQ5标记的细胞被检出。结论流式细胞术可以作为一种新的疟疾辅助诊断技术。

Objective To establish a method of flow cytometry（FCM） as a diagnostic method for malaria.Methods The blood sample of malaria patients were determined by microscope.DRAQ5 staining assay and flow cytometry were used to test malaria parasite.Results No red cell stained by DRAQ5 was detected in the healthy human blood sample（as blank control）,healthy blood sample with DRAQ5 and malaria blood sample without DRAQ5 （as negative control）,except in the Plasmodium infected blood.Conclusion The method of flow cytometry can be used as a new diagnostic method for malaria.
