肾炎Ⅱ号水丸对紫癜性肾炎小鼠血清IgA-FN含量的影响

The Effect of ShenyanⅡ pill on the Expression of IgA-FN in Blood Serum in Mice with Henoch-Schonlein purpura Nephropathy

目的:探讨肾炎Ⅱ号水丸(SY)对紫癜性肾炎(HSPN)小鼠血清IgA-纤维连接蛋白聚合物(IgA-FN)含量的影响。方法:采用口服麦胶蛋白+尾静脉注射印度墨水的方法建立小鼠紫癜性肾炎(HSPN)模型。灌胃给药17周后,ELISA法测定各组小鼠血清IgA-FN含量;免疫荧光法检测各组小鼠肾组织IgA沉积。结果:与空白组比较,模型组IgA呈团块状在系膜区沉积明显增多(P<0.01);与模型组比较,SYH组IgA沉积明显减少(P<0.01),SYM组IgA沉积明显减少(P<0.05)。与空白组比较,模型组血清IgA-FN含量明显升高(P<0.01);与模型组比较,实验各组皆可以明显降低模型小鼠血清IgA-FN的含量(P<0.01)。结论:SY降低HSPN模型血清中IgA-FN表达从而促进系膜区IgA的清除可能是其防治HSPN的机理之一。

Objective：To study the effects of ShenyanⅡpill（SY） on the expression of IgA-FN in Blood Serum in mice with Henoch-Schonlein purpura nephropathy （HSPN）. Methods：HSPN model was established by feeding gliadin and intravenous injection of Indian ink. The Sham group and Model group were administered by saline each mouse; the CI＋DI group was treated with cimetidine and dipyridamole,according to the clinical dose; The SY＋CI＋DI group was fed with cimetidine、dipyridamole and SY; the high,medium and low dose of SY groups were treated by SY in different doses. The mise were sacrificed at the end of 17 th week. The deposition of IgA in the mesangial area were observed by immunofluorescence microscope. The consentration of IgA-FN in plasma were measured by the method of ELISA. Results：Compared with the sham group,IgA deposition in model in the mesangial area incresed significantly（P〈0.01）. Compared with the model group,IgA deposition in SYH group decreased significantly（P〈0.01）,IgA deposition in SYM group decreased significantly（P〈0.05）. Compared with the sham group,IgA-FN in model in plasma increased significantly（P〈0.01）. Compared with the model group,the level of IgA-FN in plasma decreased significantly in all treated groups（P〈0.01）. Conclusion：SY can decrease the level of IgA-FN which may be the clearance receptor of IgA which can be one of the most important mechanisms of its prevention and cure of HSPN.
