犬脂肪基质干细胞体外复合珊瑚支架材料的生长特性和成骨活性 被引量:1

摘要 目的:研究犬脂肪基质干细胞(adipose derived stromal cells,ADSCs)作为种子细胞复合珊瑚体外培养的生长特性和成骨活性。方法:分离犬ADSCs,成骨诱导后复合珊瑚继续培养,以未诱导ADSCs复合物为对照,进行生长曲线、DIO染色后的激光共聚焦荧光显微镜、碱性磷酸酶(AKP)和骨钙蛋白(OCN)生物化学定量检测。结果:生长曲线示7天后ADSCs在材料上生长进入平台期,共聚焦显微镜示诱导ADSCs复合珊瑚7天后生长良好;AKP检测显示成骨诱导ADSCs-材料组在各个检测点均显著高于未诱导ADSCs-材料组(P<0.05);OCN从第7天开始表达,成骨诱导ADSCs-材料组显著高于未诱导ADSCs-材料组(P<0.05)。结论:天然珊瑚是ADSCs生长和成骨转化的良好生物支架材料。 Objective:To explore the growth character and osteogenic ability of canine ADSCs in coral scaffolds. Methods:ADSCs were isolated and cultured in osteogenic-induced and non-induced medium. The induced cells were complex with coral to co-culture,non-induced ADSCs-coral construct was served as control. The constructs were examined from growth curve,Confocal microscopy and quantitative histochemistry. Results:The growth curve demonstrated the proliferation of ADSCs on coral reached plateau after 7 days co-culture. Also Confocal microscopy and quantitative histochemistry showed ADSCs grew well and began to express osteocalcin on the coral after 7 days. Quantitative histochemistry demonstrated the higher alkaline phosphatase (AKP) and osteocalcin expression of induced ADSCs-scaffold than those of non-induced ADSCs-scaffold (P〈0.05). Conclusion:The coral scaffold is a good biomaterial for canine ADSCs growth and being induced to osteoblasts.
