摘要
目的:研究在光照条件下姜黄素诱导人胃腺癌BGC-823细胞凋亡的效应,探讨其作为光动力学治疗肿瘤新型光敏剂的可能性。方法:体外培养的胃腺癌BGC-823细胞加姜黄素后光照处理,MTT法测定细胞增殖能力,荧光显微镜和电镜观察细胞核及超微结构的改变,流式细胞术检测细胞周期分布变化。结果:姜黄素在光照条件下可显著抑制BGC-823细胞的增殖;经5μmol/L姜黄素联合光照处理24h细胞即呈现典型的凋亡形态;流式细胞术检测光照条件下姜黄素诱导BGC-823细胞周期阻滞于G0/G1期。结论:光照条件下姜黄素对人胃腺癌BGC-823细胞具有显著的凋亡诱导作用,可望进一步开发成光动力学治疗肿瘤的新型光敏剂。
Objective:To investigate the effects of curcumin with illumination on induction of apoptosis in human gastric cancer cell BGC-823. Methods:BGC-823 cell was treated by curcumin with illumination. MTT was used to measure inhibitory effects of curcumin on proliferation of BGC-823 cells. Hoechst33258 staining and TEM were conducted to determine the cell morphology. Flow cytometry was used to investigate the apoptosis and cell cycle distribution of BGC-823 cells. Results:Cell proliferation was markedly inhibited by curcumin with illumination. The typical apoptotic morphology can be seen caused by curcumin of 5μmol/L with illumination. By analysis with flow cytometry,cell cycles showed G0/G1 arrest after treatment with curcumin with illumination.Conclusion:Curcumin with illumination has the effect of inducing cell apoptosis in BGC-823 cells,suggests a potential application of photodynamic therapy for tumor.
