从卵巢形态结构及MMP-9、TIMP-1表达优选PCO大鼠造模法 被引量:1

摘要 目的优选PCO大鼠的造模方法。方法分别采用来曲唑、硫酸普拉睾酮钠、硫酸普拉睾酮钠联合HCG诱导PCO大鼠模型。应用免疫组化法检测MMP-9、TIMP-1在大鼠卵巢组织中的表达,HE染色后光镜下观察卵巢局部形态。结果 MMP-9在模型组大鼠卵巢组织中的表达增高,差异有显著统计学意义(P<0.01);TIMP-1表达在模型组大鼠卵巢组织中均降低,差异有统计学意义(P<0.01,P<0.05)。3种造模方法均能诱导PCO动物模型,其中来曲唑造模法最优,符合PCOS动物造模要求。结论采用来曲唑造模法诱导PCO大鼠动物模型效果最佳。 Objective To study the effects of different modeling on ovary histology and expression of MMP-9 and TIMP-1 in PCO rats and sift the best modeling of PCO rats. Methods Lelrozol, sodium prasterone sulfate and prasterone sulfate sodium combined with human chorionic gonadotropin were used to induce PCO rats model respectively. The expressions of MMP-9 and TIMP-1 were detected with immunohistochemical technique and the morphological changes of ovary were observed with HE staining. Results The expressions of MMP-9 in all ovary tissues in model group increased (P〈0.01) and expiession of TIMP- 1 decreased (P〈0.01,P〈0.05). All the three molding could induce the PCO models in rats, but the best method was by the lelrozol which met the requirements of the animal PCOS model.Conclusion The best modeling induced PCO rats is used by the lelrozol.
