摘要
谷氨酸(Glu)和γ-氨基丁酸(GABA)等神经递质的代谢异常是诱发多种神经系统疾病的因素之一。由于神经递质在脑内的含量很低,改良建立了一种用HPLC荧光检测Glu、GABA等4种神经递质的方法,以高丝氨酸为内标,流动相为磷酸缓冲液和甲醇,梯度洗脱20min,荧光检测。分离效果及线性关系良好,Glu、GABA的相关系数分别为0.9969、0.9996;精密度CV批内3.4%,批间5.6%;其最小检测限分别为3.3pmol、1.
Abnormal metabolism of neurotransmitters such as glutamate(Glu) and γ aminobutyric acid(GABA) might contribute to one of factors to induce the multiple central nervous system (CNS) disorders. An modified fluorometric method was established for the determination of 4 neurotransmitters such as Glu and GABA by HPLC assay. Homoserine served as internal standard,phosphate buffer solution and methanol as mobile phases, and gradient elution lasted 20 min with fluorescence detection. Separation and linear relationship were fine. Coefficients of correlation for Glu and GABA were 0.996 9 and 0.999 6, respectively. The precision was 3.4 % for interassay CV , and 5.6 % for intraassay CV . The limits of detection for Glu and GABA were 3.3 pmol and 1.6 pmol, respectively.
