摘要
从9种备选酶中选取胃蛋白酶(Pepsin)和风味蛋白酶(Flavourzyme),以脱钙的鲑鳟鱼骨制取Mr<3000Da活性胶原肽,并进行抗性测定。结果EDTA微波复合脱钙效果最佳,浓度、时间、微波功率等对脱钙效率和胶原损失影响较大;最佳酶解工艺为先Pepsin1.2%,料液比1:30,T65℃,pH2.2,提取6h,提取率达8.68%,DH2.23%,后调料液比至1:35,Flavourzyme1.2%,T57℃,pH6.4,提取4h,提取率为6.96%,DH5.81%;经粗滤、微滤、超滤、浓缩、干燥过筛等得到成品,其纯度为84.08%,羟脯氨酸(Hpro)含量为6.22%。产品FRAP值为3.80mmol/L,DPPH.清除率为13.33%,.OH清除率为11.24%。
Collagen polypeptides (Mr〈3 000 Da) were extracted from decalcifying trout bone and its anti-oxidative activity were detected.The enzymatic hydrolysis conditions for Pepsin and Flavourzyme were optimized.The results showed that the way of EDTA bonding with microwave was the best method to decalcify.Concentration of EDTA,reaction times and power of microwave were important influential factors.The best process was the ratio of material to liquid of 1:30,Pepsin added with 1.2%,temperature at 65℃,pH2.2,action for 6 hours and the yield of collagen polypeptides was 8.68% and DH was 2.23%.Then the ratio of material to liquid was changed to 1:35,Flavourzyme added with 1.2%,temperature at 57 ℃,pH6.4,action for 4 hours,and the yield of collagen polypeptides was 6.96% and DH was 5.81%.After the process of colation,microfiltration,ultrafiltration,concentration,drying and cribration,finished product with purity of 84.08% was obtained,in which the contents of Hpro was 6.22%,the value of FRAP was 3.80 mmol/L,clearance rate of DPPH· was 13.33% and clearance rate of ·OH was 11.24%.
